The former Ireland and Ulster out-half will replace David Nucifora
Nucifora will leave his role at the end of this season after a decade at the helm.
Humphreys made 72 appearances for Ireland and captained Ulster to the European Cup in 1999.
Since retiring as a player, Humphreys has been Director of Rugby for both Ulster and Gloucester and is currently the Director of Performance Operations with the England and Wales Cricket Board.
